Neurofibromatosis type 1 (NF1) is a complex genetic disorder characterized by the growth of tumors along nerves, most commonly plexiform neurofibromas (PNs), which can cause significant pain, disfigurement, and functional impairment. Historically, management of NF1-PNs relied on supportive care and surgical intervention, which offered little reprieve from debilitating symptoms. Newer targeted therapies such as MEK inhibitors (MEKi) have proven capable of slowing or shrinking PNs and alleviating symptoms, fundamentally transforming the natural history of NF1-PNs across the globe.

Children’s Brain Tumor Network (CBTN): Advancing Community-Led Discovery

Progress in pediatric neuro-oncology increasingly depends on the ability to connect data, expertise, and investigators across institutions and borders. The Children’s Brain Tumor Network (CBTN) has enabled a growing global community to contribute to and learn from shared datasets spanning molecular, clinical, and imaging domains.

This session will bring together investigators leveraging these resources to explore tumor biology, characterize disease trajectories, and identify new opportunities for therapeutic intervention. Through a series of case studies and emerging findings, speakers will highlight how collaborative, data-driven approaches are shaping current research and informing future directions in pediatric neuro-oncology.


“Targeting the Change” – From Diagnosis to Therapy in H3 K27M Diffuse Gliomas

This session, led by three experts, will encompass key advances in the understanding and management of H3 K27M diffuse gliomas. The talks will cover evolving disease biology, including tumor–neuron interactions, as well as emerging clinical strategies and therapeutic approaches.The session will also provide an overview of emerging clinical approaches, including investigational therapies such as ONC201 (dordaviprone), and discuss ongoing efforts to address the significant unmet need in this patient population.
